Blocking integrin inactivation as an anti-angiogenic therapy.

چکیده
During angiogenesis, endothelial cell migration is coordinated by integrinmediated contact with the extra-cellular matrix (ECM), coupled with receptor tyrosine kinase signalling to regulate dynamic cytoskeletal and plasma membrane reorganization. A recent paper by Vitorino et al (2015) defined a new MAP4K4– moesin–talin–b1-integrin pathway that could be therapeutically exploited to suppress pathologic angiogenesis.

Quinazoline derivative compound (11d) as a novel angiogenesis inhibitor inhibiting VEGFR2 and blocking VEGFR2-mediated Akt/mTOR /p70s6k signaling pathway
Objective(s): We previously reported a series of quinazoline derivatives as vascular-targeting anticancer agents. In this study, we investigated the mechanism underlying the anti-angiogenic activity of the quinazoline derivative compound 11d. Materials and Methods: We examined the effects of quinazoline derivative 11d on vascular endothelial growth factor receptor-2 (VEGFR2) activation via VEG...
متن کاملPotential of Integrin Inhibitors for Treating Ovarian Cancer: A Literature Review
Epithelial ovarian cancer is a fatal disease, with a cure rate of only 30%. Several recent studies have targeted integrins for cancer treatment. Preclinical studies have shown the effectiveness of several integrin inhibitors for blocking cancer progression, especially by blocking angiogenesis. Because the initial critical step in ovarian cancer metastasis is the attachment of cancer cells to th...
متن کاملIntegrin Inhibitors as a Therapeutic Agent for Ovarian Cancer
Ovarian cancer is a deadly disease, with a cure rate of only 30%. Despite aggressive treatments, relapse remains almost inevitable in patients with advanced-stage disease. In recent years, great progress has been made towards targeting integrins in cancer treatment, and clinical studies with various integrin inhibitors have demonstrated their effectiveness in blocking cancer progression. Given ...
متن کاملAcute, but not constitutive, loss of endothelial β3-integrin inhibits tumour growth and angiogenesis
Background Angiogenesis, the formation of new vessels from pre-existing ones, is essential for tumour growth and metastasis. Endothelial cells play a central role in this process: they drive blood vessel formation in response to signals from the local environment, by a mechanism that is integrindependent. We are particularly interested in understanding what role avb3-integrin plays in governing...
متن کاملThe Anti-Proliferative and Anti-Angiogenic Effect of the Methanol Extract from Brittle Star
Background: Anti-angiogenic therapy is a crucial step in cancer treatment. The discovery of new anti-angiogenic compounds from marine organisms has become an attractive concept in anti-cancer therapy. Because little data correlated to the pro- and anti-angiogenic efficacies of Ophiuroidea, which include brittle star, the current study was designed to explore the anti-angiogenic potential of bri...
